this study demonstrated that a consistent stressor can be used in all cell-based studies to determine the presence of apoptosis at any desired time. the consistent stressor, namely, p53, that was used in this study is consistent with what has been known about p53 in the research literature for over 30 years. all of the data collected in the course of this research are consistent with what has been known about p53 for over 30 years and are consistent with what is found in the published literature.there are 787 different amino acids, 15 of them have been found to be associated with human disease. of the 15 amino acids that are associated with diseases, 5 are found in proteins that are components of the apoptotic cell death pathway, namely, bcl-2, caspase-8, p53, bax and fas (fig. 1). the data presented here demonstrate that sirt1 is critical to the induction of apoptosis, not only as a core component of the apoptotic cell death pathway, but also as a downstream target of the p53 response itself.the above study demonstrated that the mutation of r446 specifically, an a at the 446 site, was able to completely prevent stac from activating the apoptotic cell death pathway. this suggests that the 446 position could be a direct target for some substance. this is confirmed by the fact that the 446 site is only 30 aa away from the stac e230 site (fig. 4b,c). since e230 is the only amino acid that is mutated in the e230k mutant, it is reasonable to conclude that in this mutant, e230 should be the only amino acid that is in the stac. as we have already demonstrated, the single charge-reversal mutation of e230k fails to act as an stac.
